I saw the "cue" you mentioned as well. But you must go back a bit further to even understand this one, I think.
Remember somewhere about halfway or so in the movie, when the Director meets with the Cowboy out on Cowboy's Ponderosa? What does the Cowboy tell him? He says, "If you do Good, you will see me one more time. If you do Bad, you will see me two times." I'm wondering if YOU remember, that was the "real" FIRST clue to the "cue" you mention, or prelude to, I think.
So, how many times was he (Cowboy) "seen"? I count the time you mentioned as one, but also the time he showed up in Diane's bedroom, when he opened the door and said, "Time to wake up, pretty lady." I think Cowboy was a Conscience of the mind. The woman who kissed Camilla Rhodes on the lips at the dinner party looked to me like the "first" Camilla we saw auditioning for the Director, when she was singing. This was "the one", or the girl he was supposed to have to select. So then after kissing her, she walks off behind the wall to the left, then Cowboy walks by quickly in the opposite direction, as if they were doing an "exchange" of sorts. So I'm sure Lynch meant some symbolism to all this as well. There are SO many bizarre things of symbolism all throughout this weird movie. It IS difficult to analyze and figure out. It's like you have to take each part of it, write it all down, and then do the best you can to piece it all together. But then when you may think you have figured one scenario out, you think about the other twists that affect that one piece you think you've figured out, and then NOTHING makes any sense (perhaps - just my opinion..)!! So, with this particular scenario of the movie, I pieced together these entities: the Cowboy, Director's choice of "the one girl", seeing the Cowboy two times (so therefore we know that it's already turning out "Bad" as Cowboy said it would if he was seen twice), and then of course he (Lynch) brings "the one" (the girl - the one the Director was forced to select from the audition) somehow into the final episodes when she stops by the table to give Rita (now Camilla) a passionate kiss. What was that supposed to be about. For sure, it hurt and made Diane jealous. I think all of these things are somehow tied into each other. But also in my opinion, I think Lynch may be one strange bird to create something like this, where it's so jumbled up that you cannot even piece all the parts together. And that makes for one f-'d up movie! I'm just wondering if anyone knows of where Lynch may have done an interview and "explained" or actually disclosed or answered the questions regarding this production? I'd sure like to see what he says!

I like this movie a lot, I've been reading all comments here and noticed that no-one saw this little clue (or perhaps it is not a clue, I don't know..).
At the dinner party close to the end of the movie, When Camilla Rhodes, after kissing Camilla exits in the back, she hurtles off in good speed and disappears behind a wall. About a second later, the Cowboy hurtles out from behind the very same wall, in same speed, in opposite direction and disappear out to the right of the picture.
Is there a connection between the Cowboy and Camilla Rhodes?
